Hi, I have a 6-day Swiss Travel Pass Youth (2nd class). Can I use it as the basis for a class upgrade (2nd → 1st class) for a specific route, like the glacier express from brig to andermatt. Thank you!

Yes, you can buy upgrades for a specific route, you pay only the Half Fare with a Swiss Travel Pass. Day Class Upgrades for 50 CHF are also available. Please note that a seat reservation is required in the Glacier Express trains, but not in the regional trains on the same route.
